Description
SJ 172550 is a small-molecule inhibitor of MDMX/MDM4 that disrupts the MDMX-p53 interaction, with reported EC50 of 2.3 µM and approximately 10-fold selectivity over MDM2. It binds the p53-binding pocket of MDMX and releases p53-dependent apoptosis signaling.
MDMX suppresses p53 tumor-suppressor function and is amplified or overexpressed in several cancers. SJ 172550 is relevant for p53 reactivation studies and for combination research with MDM2 inhibitors such as Nutlin-3a.
