Description
JNJ-55511118 is a potent negative allosteric modulator of AMPA receptors associated with TARP-γ8, with a reported Ki value of 26 nM. TARP-γ8 is enriched in hippocampal and forebrain circuits, enabling regionally biased inhibition of AMPA receptor signaling.
JNJ-55511118 shows oral activity, favorable pharmacokinetic properties and high receptor occupancy in preclinical studies. It is used to reversibly suppress TARP-γ8-containing AMPA receptors in electrophysiology, hippocampal network, seizure and excitotoxicity research while limiting broad motor effects associated with nonselective AMPA antagonists.
