Description
A83-01 is a potent inhibitor of the TGF-β type I receptor superfamily activin-like kinases ALK5, ALK4 and ALK7 with IC50 values of 12 nM, 45 nM and 7.5 nM, respectively. A-83-01 inhibits TGF-β-induced Smad signaling and epithelial-to-mesenchymal transition (EMT) while having no effect on BMP signaling.
The TGF-β/Activin pathway regulates cell differentiation, proliferation, stem cell fate, and tissue homeostasis. A-83-01 is widely used in stem cell research and has been employed to generate rat and human induced pluripotent stem cells (iPSCs) with mouse embryonic stem cell-like self-renewal characteristics. It is also a valuable tool for investigating TGF-β-mediated EMT and cellular reprogramming.

A83-01 has been reported to slowly decompose to A77-01 (Axon 1744) in solution. Freshly prepared solutions are recommended. Stock solutions in DMSO should be stored below -20 °C and used within one month.
