Description
Squarunkin A is a high-affinity inhibitor of the interaction between UNC119A and the myristoylated N-terminal peptide of Src-family kinases, with an IC50 value of 10 nM. It occupies the lipid-binding pocket of UNC119 proteins and interferes with cargo recognition.
By disrupting UNC119-dependent trafficking of myristoylated signaling proteins, Squarunkin A reduces proper localization and activation of Src kinase. It is a useful chemical probe for investigating lipidated-protein transport, UNC119 adaptor biology and spatial control of Src-family signaling.
