Description
ML 323 is a selective, reversible inhibitor of the USP1–UAF1 deubiquitinase complex with IC50 values of 76 nM in a Ub-Rho assay and 174 nM and 820 nM in orthogonal K63-linked di-ubiquitin and Ub-PCNA assays. It sensitizes cisplatin-resistant cancer cells by disrupting translesion synthesis and Fanconi anemia DNA repair pathways.
USP1–UAF1 coordinates deubiquitination events that regulate DNA damage tolerance and repair. ML 323 is valuable for studying deubiquitinase pharmacology, cisplatin sensitization and DNA repair pathway crosstalk.
