Description
NT 157 is an allosteric inhibitor of IGF1R signaling that promotes ERK-MAPK-dependent inhibitory serine phosphorylation and degradation of insulin receptor substrate 1 and 2 (IRS1/2). It shifts IGF1R complex formation from IRS1/2 toward Shc, leading to sustained inhibition of IGF1R pathway output.
IGF1R/IRS signaling supports tumor growth, survival and resistance to targeted therapies. NT 157 is relevant for studying adaptor-level IGF1R pathway suppression, IRS degradation and resistance mechanisms in oncology models.
