Description
KDU691 is an inhibitor of Plasmodium phosphatidylinositol 4-kinase (PI4K) with activity against multiple parasite liver stages. Reported IC50 values are 0.18 µM against hypnozoite forms and 0.061 µM against liver schizonts.
KDU691 also inhibits blood-stage parasites, including dihydroartemisinin-pretreated ring stages, and has demonstrated causal prophylactic activity in primate malaria models. Although it suppresses developing liver stages, in vivo studies indicate that Plasmodium PI4K inhibition alone does not reliably eliminate established dormant hypnozoites and should not be interpreted as validated radical-cure activity.
