Description

Potent Calcineurin inhibitor (calcium dependent serine/threonine protein phosphatase 2B). The mechanism of action of FK 506 (Tacrolimus) involves the formation of a molecular complex with the the immunophilin FKBP12 (IC50 value 3 nM), to reduce its peptidyl-prolyl isomerase activity. Tacrolimus potently inhibits T-cell activation-induced TNF-α, IL-1β, IL-2, IL-3, IL-4, and IL-6 production, but does not affect LPS-induced cytokine production and proliferation of normal cells, such as bone marrow cells. Immunosuppressant, neuroprotectant and anticonvulsant.
