Description
BCI hydrochloride is the hydrochloride salt of BCI (NSC 150117, Axon 2178), an allosteric inhibitor of dual-specificity phosphatases (DUSPs). BCI selectively inhibits DUSP6 and DUSP1 with IC50 values of 12.3 μM and 11.5 μM, respectively, while showing no significant inhibition of DUSP5.
DUSP6 is a key feedback regulator of fibroblast growth factor (FGF) signaling that negatively regulates ERK1/2 activation. Pharmacological inhibition of DUSP6 by BCI enhances ERK signaling and has been widely used to study developmental biology, regeneration, stem cell signaling, and cancer biology. BCI-mediated DUSP6 inhibition promotes expansion of myocardial progenitor cells and increases heart size in zebrafish embryos. In addition, BCI has demonstrated activity against lung cancer and uveal melanoma cell lines.
