Description
HAMNO (NSC111847) is a protein-interaction inhibitor of replication protein A (RPA) that selectively binds the N-terminal DBD-F domain of RPA70. It disrupts critical RPA protein interactions and inhibits ATR autophosphorylation and ATR-mediated phosphorylation of RPA32 Ser33.
RPA coordinates single-stranded DNA protection, replication stress signaling and ATR/Chk1 checkpoint activation. HAMNO is useful for studying RPA-dependent replication stress responses and cancer-cell vulnerability to oncogene-induced DNA damage.
