Description
Begacestat (GSI-953) is a highly potent and selective gamma (γ)-secretase inhibitor (GSI) that reduces Aβ40 and Aβ42 production in cells expressing human amyloid precursor protein (APP). It exhibits potency (EC50 values) of 14.8 nM and 12.4 nM for Aβ40 and Aβ42, respectively. It functions as a "Notch-sparing" inhibitor, demonstrating a >16-fold in vitro selectivity for processing APP over the Notch signaling pathway.
γ-Secretase is a multi-subunit protease complex involved in APP cleavage and Notch signaling. Begacestat is used to study γ-secretase-dependent APP processing and amyloid-β generation. Begacestat is pharmacologically relevant for Alzheimer disease research and for evaluating selective modulation of amyloidogenic processing.
