Description
Purmorphamine is a potent Hedgehog (Hh) pathway agonist that directly targets the Smoothened (SMO) transmembrane receptor. Activation of SMO by Purmorphamine stimulates canonical Hedgehog signaling and upregulates expression of key pathway mediators including PTCH1, GLI1, GLI2, and SMO.
The Hedgehog signaling pathway is a critical regulator of embryonic development, stem cell maintenance, tissue regeneration, and cell fate determination. Purmorphamine is widely used as a small-molecule substitute for Sonic Hedgehog (Shh) protein in stem cell and developmental biology applications. The compound promotes osteoblast differentiation of multipotent mesenchymal progenitor cells and lineage-committed preosteoblasts, making it a valuable tool for skeletal biology and regenerative medicine research.
