Description
ABC294640 (opaganib) is an orally active, sphingosine-competitive inhibitor of sphingosine kinase 2 (SphK2/SPHK2) with a reported Ki value of 9.8 µM. It reduces cellular formation of sphingosine-1-phosphate and shifts the balance of bioactive sphingolipids.
ABC294640 suppresses proliferation and migration in diverse cell models and shows in vivo sphingolipid-pathway activity. It also affects additional lipid enzymes, including dihydroceramide desaturase, at relevant concentrations; direct lipidomic measurement is therefore valuable when attributing phenotypes specifically to SphK2 inhibition.
