Description
A-92 is a potent GCN2 (EIF2AK4) inhibitor that suppresses the stress response mediated by general control nonderepressible 2 kinase (GCN2) with an IC50 value of <0.3 μM. A-92 inhibits GCN2-dependent signaling pathways involved in amino acid sensing and cellular adaptation to nutrient stress.
GCN2 is a key regulator of the integrated stress response (ISR) and is activated by amino acid deprivation through sensing uncharged tRNAs. GCN2-mediated phosphorylation of eIF2α promotes adaptive transcriptional programs that support cell survival under stress conditions. A-92 is a valuable tool for studying cellular stress responses, translational control, and cancer cell adaptation to nutrient limitation.
