Description
Rbin-2 (ribozinoindole-2) is a potent, reversible, and selective inhibitor of the essential AAA+ ATPase Midasin (Mdn1/Rea1), with a reported GI50 of 14 nM in wild-type cells. Rbin-2 directly inhibits Mdn1 ATPase function, preventing the ATP-dependent removal of assembly factors from pre-60S ribosomal particles.
Acute Mdn1 inhibition disrupts large-ribosomal-subunit maturation, nucleolar organization, and production of functional ribosomes without requiring genetic depletion of this essential enzyme. Rbin-2 is therefore a powerful chemical probe for temporal studies of eukaryotic ribosome biogenesis and AAA+ mechanoenzyme function.
