Description
(±)-Blebbistatin is a reversible and selective inhibitor of the motor ATPase activity of class II myosins. Reported IC50 values for inhibition of actin-activated ATPase activity are 6.47 µM for smooth-muscle myosin, 3.58 µM for nonmuscle myosin IIA, 2.30 µM for nonmuscle myosin IIB and 1.57 µM for nonmuscle myosin IIC.
Blebbistatin preferentially stabilizes a myosin–ADP–phosphate intermediate, slowing phosphate release and suppressing force generation without directly inhibiting myosin light-chain kinase. It is widely used to study actomyosin contractility, cytokinesis, cell migration and mechanobiology; light exposure should be minimized because the parent scaffold is photolabile and can produce phototoxic effects.
