Description

Lalistat 2 is a potent inhibitor of lysosomal acid lipase (LAL/LIPA) with an IC50 value of 152 nM. It shows little inhibition of human pancreatic lipase or bovine milk lipoprotein lipase at concentrations used in the original selectivity studies.

Lalistat 2 is widely used to distinguish lysosomal acid lipase activity from neutral lipase activity in biochemical and dried-blood-spot assays. It is also used to model impaired lysosomal cholesteryl-ester and triglyceride hydrolysis, although reported effects on neutral lipid metabolism should be considered when interpreting prolonged cellular exposure.

Key Features

  • Potent LAL inhibitor
  • IC50: 152 nM
  • Useful for differential LAL activity assays
  • Limited activity against pancreatic and lipoprotein lipases in initial profiling

Applications

  • LAL and LIPA enzyme assays
  • Dried-blood-spot lipase measurements
  • Lysosomal lipid-metabolism studies
  • Cholesteryl-ester hydrolysis research
  • Lysosomal acid lipase deficiency models
