Description
SMU-L11-R is a potent and selective Toll-like receptor 7 (TLR7) agonist with an EC50 value of 12 nM against human TLR7. As the biologically active R-(-)-enantiomer of racemic SMU-L11, SMU-L11-R potently activates TLR7-mediated innate immune signaling.
Activation of TLR7 by SMU-L11-R stimulates NF-κB and MAPK signaling pathways, resulting in increased production of pro-inflammatory cytokines including TNF-α and IL-6. In preclinical MC38 colorectal cancer models, combination treatment with SMU-L11-R and PD-L1 checkpoint inhibitors produced a robust antitumor response through activation of both innate and adaptive immunity. Treatment increased infiltration and activation of natural killer (NK) cells, macrophages, dendritic cells, and cytotoxic CD8+ T lymphocytes, leading to enhanced tumor control in vivo.
