Description
UNC3474 maleate is a 53BP1 antagonist with an IC50 value of 14 µM and a Kd value of 1 µM. UNC3474 targets the methyl-lysine binding cavity in 53BP1TT function by stabilizing a previously unknown lowly populated autoinhibited homodimeric state of 53BP1TT in which the histone binding surface of H4K20me2 is buried within the protomeric interface.
