Description
RBPJ inhibitor RIN1 is a potent, selective and first-in-class RBPJ transcription factor inhibitor. RIN1 inhibits RBPJ in both its activating NOTCH complexes and inhibitory SHARP complexes, disrupting RBPJ-dependent transcriptional regulation.
RBPJ, also known as CBF1, KBF2 or CSL, is the central DNA-binding transcription factor of canonical NOTCH signaling. RBPJ integrates activating and repressive protein complexes to regulate gene expression programs involved in cell fate, differentiation, proliferation and cancer biology. Consistent with disruption of NOTCH signaling, RIN1 inhibits proliferation of hematologic cancer cell lines and promotes skeletal muscle differentiation from C2C12 myoblasts.
