Description
ML346 (CID 767276) is a cell-active activator of the heat-shock response and Hsp70 expression, with an EC50 of 4.6 µM in HeLa cells. It induces HSF1-dependent expression of molecular chaperones, including Hsp70, Hsp40, and Hsp27, and also engages broader proteostasis programs involving oxidative-stress and unfolded-protein-response pathways.
ML346 restores protein folding and function in cellular models of conformational disease, including rescue of mutant CFTR-dependent ion transport, without significant cytotoxicity at active concentrations. It is useful for studying pharmacological reinforcement of the proteostasis network, HSF1-regulated chaperone expression, and the capacity of stress-response pathways to reduce misfolded-protein burden.
