Description
PY-60 is a robust and selective activator of YAP (Yes-associated protein) transcriptional activity that acts by targeting annexin A2 (ANXA2), with a Kd value of 1.4 μM. PY-60 promotes YAP activation through modulation of ANXA2-dependent signaling, resulting in increased levels of transcriptionally active YAP.
The Hippo-YAP signaling pathway is a central regulator of cell proliferation, tissue regeneration, stem cell function, organ growth, and wound repair. PY-60 treatment liberates annexin A2 from the plasma membrane, ultimately promoting a phosphatase-bound, non-phosphorylated, and transcriptionally active form of YAP. As a result, PY-60 provides a valuable chemical tool for studying YAP-dependent transcription, Hippo pathway regulation, regenerative biology, and tissue repair mechanisms.
