Description
ML 154 (NCGC00185684) is a competitive, selective and brain-penetrant neuropeptide S receptor (NPSR) antagonist with IC50 values of 22.1 nM, 36.5 nM and 5.0 nM in Ca2+, cAMP and binding assays, respectively. It preferentially blocks NPS-induced ERK phosphorylation and decreases alcohol self-administration in vivo.
NPSR signaling regulates arousal, anxiety-like behavior, reward circuitry and alcohol-seeking behavior through GPCR-linked pathways. ML 154 is useful for studying NPSR antagonist pharmacology and addiction-related neurobehavioral mechanisms.
