GAT211 is a potent CB1 receptor ago-PAM with allosteric agonist activity residing exclusively in the R-(+)-enantiomer, GAT228 (Axon 4358), and S-(-)-enantiomer, GAT229 (Axon 4359), acting as a “pure” and potent CB1R PAM. GAT211 engages CB1R allosteric site(s), enhances the binding of the orthosteric full agonist [3H]CP55,490, and reduces the binding of the orthosteric antagonist/inverse agonist [3H]SR141716A.
